The experimental technique consists in physically and virtually screening the efficacy and the toxicity of the substances, in vitro and in vivo. The best objective of the A-PARADDISE project is to supply several candidate treatments against the four parasites and to pave the way for clinical trials soon. To ensure the success of the project, the individuals were all selected for their high level of expertise within their respective areas: high-throughput screening, computer-aided screening, the production of recombinant proteins, following generation sequencing, phenotypic exams, toxicology and pharmacology..The study found a significant increase in the expression of the enzyme and creation of 12-HETE in humans with advanced-stage diabetic retinopathy and in pets with induced ischemic retinopathy. Al-Shabrawey treated retinal M-ller cells in vitro with 12-HETE also. He found this improved the amount of vascular endothelial growth factor and decreased pigment epithelium derived aspect, disrupting the balance and resulting in neovascularization. By using this finding, Al-Shabrawey’s group discovered that treating the animals with an enzyme inhibitor or inducing ischemic retinopathy in pets lacking the enzyme restored that balance and inhibited neovascularization.
